ラットに逆行性神経トレーサーであるFluorogold(FG)を右足関節内に1,1’dioctadecyl-3,3,3’,3’-tetramethyl-indocarbocyanine perchlorate (DiI)を右ヒラメ筋膜下に注入し、7日後に蛍光顕微鏡下に足関節とヒラメ筋に分布する髄節・神経根レベルおよび血管周囲神経を確認した。次に、ラットでの関節拘縮・筋萎縮・関節症モデルにおいてvon Frey testによる痛覚過敏の評価法を確立した。本モデルでは、損傷1週間より、痛覚過敏が誘発され、さらに3週間以降も、痛覚過敏が増強した。続いて、若年ラット（8週齢）を用いた本モデルに対しストレッチ、運動療法、温熱療法など各種理学療法手技を加え、von Frey testにて測定した痛覚閾値と神経・筋・関節組織における痛覚関連物質の発現パターンとの相関を検討した。痛覚関連物質として、カルシトニン遺伝子関連ペプチド(calcitonin gene-related peptide, CGRP)、サブスタンスP（Sub-P）、グルタミン酸受容体、グルタミン酸トランスポーター、血管作動性腸ペプチド（vasoactive intestinal peptide, VIP）、一酸化窒素合成酵素 (NOS)、ソマトスタチン、ニューロペプチドY、インターロイキン1β (IL-1β)、腫瘍壊死因子α (TNFα)、インターフェロンγ (IFNγ)、Macrophage inflammatory p rotein-1α (MIP-1α)、Nerve Growth Factor（NGF）をRT-PCR、Western blotting、免疫組織学にて測定した。これらのうち、 CGRP、Sub-P、NGFをラットの下肢関節・筋・血管周囲神経に認めていたが、理学療法手技による痛覚閾値の変化と有意な相関を示したものはなかった。そのため、新規疼痛関連物質を検索したところ、神経伸長阻害因子の一つであるSemaphorin 3Aの発現を関節周囲組織に認め、その分布、発現量について検討した。関節拘縮・筋萎縮・関節症の進行また加齢に伴い、関節周囲組織でのSemaphorin 3Aの発現減弱を認め、痛覚閾値の低下との関連が示唆される結果を得た。
