哺乳動物の中枢神経系では発生初期で過剰に生産された神経細胞がプログラム細胞死を来すため、生後発達過程において神経細胞数は減少することが一般に知られているが、その経時的な変化の詳細や神経系の機能発達との関連は未だ明快に解明されていない。そこで、本研究では脳組織薄切切片内における神経細胞核の形状に着目した新定量法を確立し、それを用いて支配筋の運動機能発現時期が異なるラットの顔面運動神経細胞数を経時的に定量し、生後発達に伴うプログラム神経細胞死に起因する経時的な細胞数減少を明らかにした。その定量結果とそれぞれの顔面筋の運動機能発現時期から、顔面筋の運動機能発現以前に比べてそれ以降では明らかに顔面神経細胞数の減少率が低下していることが判明した。このことから、プログラム神経細胞死に起因する顔面神経細胞数の減少は、顔面筋の運動機能の発現と密接に関連していることが明らかになった。また、今回利用した新定量法は高い正確性を有することが確認されたため、今後の細胞数定量におけるStandardとなることが期待される。
