日本でも近年患者数が増加し、社会問題となっている、アルツハイマー病のモデル動物を作成し、種々のRIリガンドを用いたオートラジオグラフィを比較検討することにより、ECTイメージングによる臨床的評価の基礎的資料を提供する目的で本研究を行った。前脳基底部をイボテン酸で破壊したラットに自家副交感神経である迷走神経節を大脳皮質に移植し、step-through型受動的回避実験装置を用いた受動的回避学習の反応保持テスト、組織化学的検索並びにコリンアセチル転移酵素及びアセチルコリンエステラーゼ活性にて移植が生着することを確認した。破壊手術群、シャム手術群、移植群において、^3H-QNBを用いたmAChRインビトロオートラジングラフィ、^<35>S標識オリゴヌクエオチドプローブを用いたm1,m2mAChR-mRNAのin situ hybridization、^3H-Vesamicol及び^3H-Hemicholiniumを用いたACh Transporterインビトロオートラジオグラフィを行った。その結果、1.^3H-QNBでは3群ともに有意な変化を認めなかった。2.m1mAChR-mRNAでは破壊群において2週間で増加、4週間で減少した。一方,移植群では2週間で減少,4週間で増加した。3.m2mAChR-mRNAではm1mAChR-mRNAと逆の結果を示した。4.^3H-Vesamicol及び^3H-Hemicholiniumでは破壊群で減少した。移植群の例数が少なく更に検討を要するが、移植効果判定に有効であると考えられた。Hemicholiniumは脳血液関門を通過しないので、VesamicolをRIで標識した方がECT用として有望であり現在^<125>I標識Vesamicolについて検討中である。
