1.目的:in vitroにおける、リポ蛋白添加によるヒト単核球あるいは単球の凝固活性化について検討したので報告する。2.方法:種々の濃度VLDL、LDL、HDLを、単核球、単球、リンパ球にそれぞれ添加し、37℃、5%CO_2下で培養し、浮遊液のprocoagulant activity(PCA)、tissue factor(TF)-like activity、TF抗原量を測定した。3.成績:(1)単核球の検討:単核球(単球20%、リンパ球80%の混合)にVLDLを添加した浮遊液では、濃度依存的にPCA、TF-like activity、TF抗原量は増加し、100μg/ml VLDLの添加にてTF-like activityは対照の約20倍、TF抗原量は対照では感度以下であったのに対し、762±79pg/10^5cellまで著増した。LDL、HDLを添加した場合、低濃度では対照と有意差を認めなかったが、100μg/mlの濃度にて対照と有意差を認めた。(2)単球の検討:単球にVLDLを1μg/ml、50μg/mlと各濃度を加えて培養し、浮遊液中のPCA、TF-like activityを測定すると、対照と比較して有意に上昇していた。(3)リンパ球の検討:リンパ球に種々のリポ蛋白を添加した浮遊液では、PCA、TF-like activity、TF抗原量は対照と有意差を認めなかった。4.考察:今回の検討より、VLDLは単球を活性化し、TF産生を促進すると考えられ、高脂血症患者における凝固亢進状態を引き起こす要因のひとつになるかもしれない。リンパ球にリポ蛋白を添加した場合にはTF-like activityを認めず、リンパ球と単球の混在した場合に、より高いTF-like activityを認める可能性が示唆され、この点については今後さらに検討を要する。
