C3H系マウスにネンブタールの腹腔内投与による麻酔を施行後、軟X線装置にて250kV、20mAで背部皮膚に放射線照射を行った。照射量は10,20,30Gyに設定した。背部皮膚のパラフィン切片を作成し、H・E染色、エラスチカ・ワンギーソン染色で組織学的変化を観察した。その結果、照射量が増加するに従って皮膚付属器(毛根)の変性、脱落、消失が著しく観察された。表皮の細胞層は減少し、その萎縮が観察された。また、上記モデルを用いて照射1週、4週、12週目におけるNotch受容体(Notch-1)、そのリガンド(Jagged-1)、シクロオキシゲナーゼ(COX-1、COX-2)の発現を免疫組織学的に検討した。Notch受容体はリガンドと結合することにより細胞の分化や維持に関わっている。Notch-1のリガンドであるJagged-1は分化を抑制するシグナルを活性化し、広範な細胞の遊走を促進する。COX-1は生体内に恒常的に発現しているが、COX-2はストレスや炎症により発現が誘導される。照射1週間目ではNotch-1とJagged-1が毛根付近に強く発現していた。COX-1とCOX-2の発現は弱かった。4週が経過して放射線照射からの治癒機転が始まる頃になると、Jagged-1は毛根付近に限局したままであったがNotch-1はより表層の角化層まで発現が拡大した。COX-1とCOX-2はともに間質の線維芽細胞に強く発現していた。12週目では組織全体の線維化が進み、これらの物質の発現は低下していた。本研究から、皮膚組織において放射線照射後早期に炎症などの強い障害により変性したのち、時間とともに炎症が沈静化し組織の線維化が進行していく過程が明らかとなった。
Twenty C3H mice were anesthetized by intraperitoneal injection of Nembutal, and irradiated to dorsum skin (each 10 mice to 10Gy, 20Gy, 30Gy). To analyze the histological change, paraffin sections of dorsum skin were prepared for Hematokxylin and Eosin stain and Elastica van Gieson stain. As a result, the degeneration, defluvium, and disappearance of hair root was observed. In epidermis the decrease of cell layer and the atrophy were observed.The expression of Notch receptor (Notch-1), ligand of Notch receptor (Jagged-1), cycloxygenase (COX-1, and COX-2) were immunohistologically analyzed at post-irradiation day 7, 30, and 90. Notch receptor regulates the differentiation and maintenance of cells by binding to its ligand. Jagged-1 activates the signals suppressing the differentiation and accelerates the migration of various cells. COX-1 homeostatically exist in vivo, whereas the expression of COX-2 was induced by stress, inflammation, and so on. At post-irradiation day 7, the expression of Notch-1 and Jagged was strongly recognized by hair root.The expression of COX-1 and COX-2 was less observed. At post-irradiation day 30, the expression of Jagged-1 was locally observed by hair root, whereas the expression of Notch-1 extended to stratum corneum. The expression of COX-1 and COX-2 was strongly observed in fibroblasts. At post-irradiation day 90, fibrous degeneration of dorsum skin was markedly recognized and the expression of these substances was decreased.This study demonstrates that tissue is degenerated by inflammation due to irradiation at early post-irradiation time, and then inflammation gradually decreases and fibrous degeneration proceeds.
