重症複合免疫不全症(SCID)マウスのleaky現象の原因遺伝子(群)のクローニングを最終的な目的として、高率にleaky現象を発生する系統を選抜育種により確立することを試みた。用いる系統としてはC.B-17 scid/scidマウスにKSN-bgマウスを交配して得たF2マウスから血清中のIgMの量を指標にscid/scidのマウスを選抜し、以後血中のIgM量が検出限界以下の個体を選抜して数代の兄妹交配を続けた。得られたマウスのleaky出現率は生後約2ヶ月齢で20〜30%である。これらのマウスから、一方ではleaky現象の見られない個体同士の交配を続けると共に(non-leaky選抜群)、leakyマウス同士の交配による選抜を開始した(leaky選抜群)。尚、leakyの判定は血清中あるいは腹腔洗浄液中のIgM含量をELISA法により測定し、血中濃度1μg/ml以上、腹腔洗浄液中濃度、0.01μg/ml以上を示したものについてleakyマウスと判定した。測定に用いられたマウスは、non-leaky選抜群 '94年9月〜'95年3月 25litter '95年4月〜'95年10月 26litterleaky選抜群 '94年9月〜'95年3月 27litter '95年4月〜'95年10月 35litterであった。leaky出現率は、non-leaky選抜群 '94年9月〜'95年3月 24.5±24.8% '95年4月〜'95年10月 19.8±22.6%leaky選抜群 '94年9月〜'95年3月 38.3±27.0% '95年4月〜'95年10月 47.3±23.4%であり、non-leaky選抜群では明らかにleaky出現率が上昇し、一方、leaky選抜群ではleaky出現率が低下する傾向が見られた。leaky現象を支配する遺伝子(群)の存在が強く示唆された。
