交換神経節細胞等でアセチルコリンはムスカリン性受容体を刺激し、緩除な興奮性シナプス(slow EPSP)を生じる。我々は、ムスカリン性受容体刺激によりMと呼ばれる膜電位依存性カリウム(K)チャンネルの制御機構について、研究してきたが、正確な制御メカニズムが未だ理解されていない。Mカリウムチャンネルの制御機構を知る目的を果たすために、Kチャンネルの遺伝子をとり、アミノ酸配列により、考えられるイオンポアの特性を推定し、制御機構例えばどういうキナーゼによるリン酸化があり得るかを推定することにした。RANソースとしてニューロブラストーマNG108-15細胞を培養した。RNAをアフリカツメガエカル卵母細胞に分注し、2〜5日培養する。膜電位を-80mVに固定し、約1分脱分極刺激を加え、Mチャンネルの開口によるる外向き電流を記録した。あるいは電位を-30mVに固定し-60mVへのステップをかけ、Mチャンネルの閉口による内向き電流を測定した。その結果、Mチャンネルをコードするクローンをひろうことは難しいことが判明した。そこで、抑制クローニングを用いることとし、小脳よりンチャネルのような不活化しないでんりゅうに関与するクローンを拾った。そのcDNAは、474個のアミノ酸をコードするもので、膜を12回貫通する構造をもつことがわかった。酵母や線虫のゲノム計画でみつかったももと20-35%の相同性をもっていたが、ほ乳類では知られていない新規のたんぱく質であった。このたんぱく質を卵母細胞に発現したところ、それ自身では電流を通さなかった。今後このたんぱく質の性質を詳しく調べるつもりである。
