この研究では、遺伝子改変を施した精子あるいは生殖可能な精子形成細胞を作りだし、それを利用して効率よく哺乳動物の形質転換を行う方法を開発することが目的である。今年度の研究では、精子分化途中の段階にある精子形成細胞へ遺伝子を導入し、導入された遺伝子の発現を解析した。まず、研究代表者らが以前に開発したラット精子形成細胞の初代培養系を利用して、精子分化のさまざまな段階にある精子形成細胞を分取した。これらの細胞群に、エレクトロポレーション法およびリポフェクション法によリ、サイトメガロウイルスプロモーターの制御下にグリーンフルオレッセントプロティン(GFP)を発現するプラスミドDNAを導入し、GFPの生産を蛍光顕微鏡を用いて検討した。しかし、すべての精子形成細胞群について、いずれのDNA導入法を用いても、はっきリしたGFPの発現は観察されなかった。そこで次に、DNAを導入した精子形成細胞をセルトリ細胞と共培養した後にGFPの発現を調べたが、この場合も結果はネガティブであった。これらの結果は、精子形成細胞へのDNAの導入効率が低いのか、あるいは導入されたDNAからの遺伝子発現が弱いかのどちらかによると思われる。前者については、ウイルスベクターの利用が有効だと思われるが、現段階では有効なベクターの候補が見当たらない。後者については、精子形成細胞内で効率よく遺伝子転写を導くプロモーターを見いだし、それを導入ベクターに取リ入れる工夫が必要である。
