これまでに多くの医薬品のリード化合物が天然から得られているが、放線菌を初めとする菌類の培養液や生薬などの植物に加え、近年、海洋生物から得られた化合物をリード化合物とするものが次々と臨床試験に入っており、他にも多くの海洋生物由来の化合物が前臨床試験中である。中でも、カリブ海原産のホヤから単離されたエクチナサイジン743は、抗がん剤として実用化されている。このように海洋生物からは特異な構造を有する生物活性物質が多く発見されている。しかし、海洋生物由来の化合物探索研究の歴史は浅く、1960年代後半にカリブ海産腔腸動物ヤギから大裏のプロスタグランジンが発見されたのが初めての海洋由来の生物活性物質であり、伝承薬用植物に比較して、生物活性が調べられているものは意外なほど少ない。そこで、本研究課題では、生物種の豊富なカリブ海における海洋生物をターゲットとして含有成分の生物活性を広く調査し、医薬品のシーズ探索の端緒とすることを目的としている。また、海外共同研究者の清水譲教授は赤潮発生の原因となっている渦鞭毛藻の生物学・化学の第一人者である。渦鞭毛藻からはブレベトキシンなどイオンチャネルへ作用するポリエーテル化合物が多く単離されているが、本研究を行なうにあたり、カリブ海域だけでなくアメリカ北大西洋沿岸における渦鞭毛藻類の調査も広範に行ない、各種生物活性を有する化合物を探索するための材料としての海洋生物の調査を行なうことを計画した。平成15年度は、米国ロードアイランド大学薬学部の清水譲教授のもとで今後の研究に関する打合せを行なった。そして、平成16年度には、米国自治領プエルトリコのカリブ海沿岸において無脊椎生物など海洋生物の採集を行なった。
The marine environment has proven to be a rich source of extremely potent compounds that have demonstrated significant activities in antitumor, antiinflammatory, analgesia, immunomodulation, allergy, and antiviral assays. Especially, the marine invertebrates in the Caribbean afforded the variety of compounds. Among them ecteinascidin 743 was known to be an antitumor compound. In this project we collected marine invertebrates in the Caribbean and prepared samples for screening of drug discovery.
