骨粗しょう症診断への適用を究極の目的としてビタミンD第2相代謝物を合成し,それを指標として生体内における存否を明らかとした.ついてその定量法を開発し,実試料への適用を試みた.まず,各種抱合型代謝物の中でも主要なものと推測される25-hydroxyvitamin D_3[25(OH)D_3]及び25(OH)D_2各々のsulfate(S体),glucuronide(G体)を以下のルートで合成した.すなわち,G体はいずれも対応する原料より約20工程を経てプロ体を合成し,これをKoenigs-Knorr反応に付し,G体となした.なお,目的物には3位及び25位の位置異性体が存在するため,25位又は3位を事前に保護したプロ体を基質とした.成績体を各々光,熱異性化反応に付し9,10-セコステロイドへ導き,その後,保護基を除去して目的とするG体,計4種を得た.S体は対応するD体又は3位を保護したD体をスルホン化し,HPLC等で精製した.いずれの構造も各種機器データや酵素水解の知見を基に確認した.上記の方法で得られた標品を用いてHPLCによる位置異性体相互の分離を検討し,満足すべき結果を得た.さらに,先に開発したCookson型プレラベル化剤であるMBOTADによる誘導体化を行い分離の改善と高感度化を達成した.これらの知見を基にその存在が疑問視されていたヒト血中25(OH)D_33Sの存在を明らかとし,UV検出HPLC法による定量法を確立した.さらに,本法を腎不全患者血中濃度測定に適用し,健常人のそれに比し著しい低値を示す場合のあることを見い出した.また,G体の標品を用い胆管ろうラットにおいて3位又は25位にglucuronidationされることを示唆する知見を得た.
The measurements of the serum/plasma concentration of vitamin D metabolites, especially 25-hydroxy-and 1alpha, 25-dihydroxy-vitamin D_3, are videly used as a means of assessing the vitamin D status in man, especially diagnosis of osteoporosis. However, the occurrence and significance of conjugated metabolites of vitamin D in biological fluids are still poorly understood owing to the lack of authentic samples. According to the above data, the following works have been done.1. The syntheses of monoglucuronides and sulfates of 25-hydroxyvitamin-D_2 or -D_3 have been done.2. The occurrence of 25-hydroxyvitamin D_3 3-sulfate in human plasma has been clarified.3. The determination method of 25-hydroxyvitamin D_3 and its sulfate has been established and the method has been applied to the determination of these metabolites in human plasma.4. Vitamin D_3-and 25-hydroxyvitamin D_3-monoglucuronides have been identified from biliary metabolites obtained from rats dosed with vitamin D_3 and 25-hydroxyvitamin D_3, respectively.
