マトリックスメタロプロテアーゼ(MMP)には、分泌型MMPと細胞膜貫通型MMP(MT-MMP)が知られている。本研究では、細胞膜表面において細胞外マトリックス(ECM)分解に主役を演じるMT-MMPの生化学的性質とヒト病的組織における発現を調べた。得られた主な結果は、以下の如くである。1.細胞膜貫通ドメインを欠失した△MT3-MMPをE.coliで発現し、その可溶性画分から△MT3-MMPを精製した。△MT3-MMPはゼラチン、プロテオグリカン、フィブロネクチン、ヴィトロネクチンを分解し、II型とIII型コラーゲンを3/4と1/4のフラグメントに切断した。また、I型コラーゲンのテロペプチドを分解した。これらのデータから、MT3-MMPはMT1-MMPとよく似た基質特異性をもつECM分解酵素であることが明らかとなった。2.ヒト変形性関節症と慢性関節リウマチの関節軟骨と滑膜においては、MT1-MMPとMT3-MMPが発現されており、潜在型MMP-2の活性化に関与することが示された。3.ヒト乳癌と甲状腺癌組織においては、MT1-MMPは検索した全ての症例で強く発現され、潜在型MMP-2活性化と相関した。一方、MT2-MMPは約25%の症例で陽性であり、MT3-MMPの発現は検出されなかった。4.MT1-MMP遺伝子導入腫瘍細胞をマウス尾静脈内注入すると、血液中の潜在型MMP-2の活性化により転移能昂進を示した。以上のデータから、MT1-MMPとMT3-MMPは、潜在型MMP-2の活性化と自分自身のECM分解活性により、癌や炎症性組織における細胞膜表面でのECM分解を通して組織破壊に関与することが示唆された。
