培養細胞には金沢大学癌研究所化学療法部から供与されたmurine melanoma cell,B16/F10を用いた。C57/BL/6Nマウスの6-8週齢の雌をネンブタールにて麻酔し、27ゲージの針にて、2X10^6個の細胞を左心室に注入した。腫瘍細胞注入後、1週目、2週目に各5匹づつ屠殺し、全身灌流後、10%ホルマリン液にて灌流固定した。脱灰後HE染色にて脊椎正中矢状面の切片を顕微鏡下に観察した。同種正常マウス5匹を対照に用いた。細胞注入後、1週目には、5匹中1匹(5椎骨)に脊椎転移巣を認めた。腫瘍細胞の局在は、椎体上下の軟骨板に接する部位を中心に存在していた。しかし、椎体から脊柱管に腫瘍細胞が進展している像は観察されなかった。2週目においては、病理学的には5匹全例に脊椎転移巣を認め、合計52椎骨、1匹平均10.4椎骨に転移腫瘍細胞が存在していた。また5匹中1匹に下肢麻痺が生じた。21椎骨において、転移腫瘍細胞が硬膜外腔に進展し、脊髄前方、後方、もしくはその両側より脊髄を圧迫していた。硬膜外腔での上下方向の腫瘍進展範囲は1椎体以内のものが10部位、1椎体以上2椎体以内のものが10部位、4椎体に及んだものが1部位であった。このなかで4椎体のレベルにまで上下方向に腫瘍進展を認めた1部位で、腫瘍細胞は硬膜を破壊し脊髄内に直接浸潤していた。しかし他の20部位において硬膜は腫瘍の進展を完全に阻止し、脊髄内への直接浸潤は認めなかった。左心室に腫瘍細胞を注入することにより、脊椎転移モデルの作製が可能であった。特に硬膜外腔に進展した腫瘍細胞は硬膜外腔を上下方向に進展するが、硬膜内浸潤は稀であり、硬膜がバリアー効果を発揮していたことが観察された。しかし範囲が多椎体(4椎体以上)に及ぶときは、腫瘍が硬膜を破壊し、硬膜外直接浸潤の可能性もあることが証明された。
