平成7年度,我々は,NIH/3T3細胞をKi‐rasでトランスフォームしたDT細胞が,増殖因子によりCa^<2+>オシレーションを生じるメカニズムについて研究した.細胞内Ca^<2+>オシレーションは,細胞外のシグナルをオシレーションという持続的で,かつパルス状の細胞内のシグナルに変え,GTP結合タンパク質にカップルする受容体の情報の増幅を行なう重要な細胞現象である.細胞外からの,Ca^<2+>流入が,この現象を維持するため重要であることが指摘されているので,外側からの流入を実測する目的で,膜電流測定装置(既存)とCa濃度測定装置をドッキングさせ研究した.本年度は,流入が生じていることをMn^<2+>を用いて蛍光量の減少(Mnクエンチ)で示した.また,この流入機構にras‐GTPase活性とマップキナーゼの活性化が必要であることを示すデーターを得た.イギリスの研究グループが,イノシトール四リン酸と結合タンパク質が,ras‐GAPであることを示したが,それには本研究が少なからず影響を与えている.
