既存のような静的な抗がん剤ではなく、反応性を持つ動的な抗がん剤として我々が開発した抗がん化合物であるCNDACを母化合物とし、4位のアミノ基にアシル基を導入した誘導体を合成し、M5076肉腫を移植したマウスの生存率および腫瘍抑制率によって抗腫瘍効果を評価した。BD2F1マウスにM5076肉腫を皮下に移植し、3日おきに計6回経口投与を行い、20日後の腫瘍抑制率および60日後の生存率を求めた結果、アシル基としてC=16のパルミチン酸を導入したPal‐CNDACが腫瘍抑制率99.2%、生存率5/6と母化合物よりも優れた抗腫瘍効果を示した。次にヒト腫瘍に対する効果を検討する目的で24種のヒト腫瘍培養細胞を用いて、in vitroにおける殺細胞効果を検討した結果、大腸がん、乳がんのそれぞれ1株を除いたすべての細胞株で母化合物よりも強い殺細胞効果を示した。また母化合物ではほとんど効果を示さない膵がん、乳がんに対しても有効であった。次にin vivoにおける抗腫瘍効果をヌードマウスに皮下移植した胃がん(4株)、乳がん(3株)、肺がん(1株)を用いて検討した。移植後、腫瘍サイズが100‐300mm^3に達したところで薬剤を経口投与で3日おきに計6回投与したところ、フルオロウラシル系の核酸代謝拮抗剤が効きにくい胃がんのSt‐40やSt‐4に対して、Pal‐CNDACは母化合物のCNDAC、5'‐DFUR、5‐Fuなどよりも少ない投与量(150mg/kg)で95%以上の高い腫瘍抑制率を示した。この様にin vitro、in vivoそれぞれにおいてヒト腫瘍に対し、CNDACにアシル基を導入したPal‐CNDACは強い抗腫瘍効果を示し、臨床応用が期待される。
