Clostridium difficile KZ 1647株は合成培地(6xMADM)中のビオチン濃度が低いと(0.05 nM)、菌の増殖は抑制されるが、毒素(トキシンAおよびB)産生が極めて増強される(ビオチン効果)。この毒素産生増強の調節機構に関し以下の知見を得た。(1)低濃度ビオチン培地と高濃度ビオチン(50 nM)培地における菌体外タンパクを比較すると、低濃度ビオチン培地特有のタンパクとしては毒素タンパク以外には特になく、ビオチン効果は菌体外タンパクの産生を広範に増強するものではなかった。(2)低濃度ビオチン培地で、各培養時期に高濃度ビオチンを添加し、ビオチン効果への影響を検討した。毒素合成の初期段階までに高濃度ビオチンを添加すると、増殖抑制は解除されたが、毒素合成の増強は完全に抑制された。毒素合成の開始後に高濃度ビオチンを添加すると、増殖抑制はかなり解除されたが、毒素合成の増強は抑制されなかった。このことは、低濃度ビオチン状態が毒素の合成初期まで必要であり、一旦、毒素合成が開始されると、高濃度ビオチンを添加しても毒素合成の増強は不可逆的に進行する傾向にあることを示している。(3)ビオチン効果は、特定のアミノ酸(リシン、グルタミン酸、グルタミン、アスパラギン酸)によって強い抑制を受けた。ビオチン結合酵素はアミノ酸代謝に関与するので、低濃度ビオチン状態は、アミノ酸代謝系のストレス状態を起こすと考えられる。この様なストレス状態が、毒素合成の増強を誘導する可能性が示唆された。上記アミノ酸が培地中に存在すると、ストレス状態を緩和して、毒素の合成は抑制されたと思われる。(4)菌体より抽出したRNAに対し、ノザンハイブリダイゼイション法により、毒素遺伝子のmRNAの検出を行なった。2種の毒素遺伝子のmRNAは、低濃度ビオチン状態で毒素合成時期に一致して検出された。毒素遺伝子の転写レベルでの活性化が、毒素合成の増強の原因であるかは、今後検討する予定である。
