肝組織および肝外のC型肝炎ウイルスHCVの感染ならびに増殖部位として疑われる末梢血単核球を対象としてHCV関連抗原の検出を蛍光抗体法にて、また、HCVRNAをFISH(fluorescent in situ hybridization)法にて検出し、共焦点レーザー走査顕微鏡を用いて、陽性細胞の同定ならびに細胞内局在の検討を行った。しかし、組織におけるRNAの保存状態が不安定なため、安定した検出は困難であった。そこで、検出感度の改善を目的として、polymerase chain reaction(PCR)法を組織におけるin situ hybridization(ISH)法に応用したISPCR法を試みた。しかし、この手法でも組織上でHCVRNAの増幅と検出は一部の組織において可能であったが、再現性よく検出することは困難であった。HCVRNAを組織上で再現性よく検出するためにはISPCR法による増幅は必須と考えたが、現時点では、陽性対照が得られていない。そこで、肝生検組織での標的核酸の増幅の程度を評価するために、DNAウイルスであるB型肝炎ウイルス(HBV)を対象として、安定した手技の開発を行った。HBVはヒト肝臓に感染するDNAウイルスであり、その感染ウイルス量はHCVの約1000倍と報告されている。さらにDNAウイルスであるため増幅の際のreverse taranscriptionの段階を必要とせず、DNA以降の段階における手法を検討することができる。そこで、HBV及びその変異株に対するprobeならびにprimerを作成し、組織上にて、ISH法ならびにPrimerによるone step検出法であるprimed in situ labelling(PRINS)法による検出を行い共焦点レーザー走査顕微鏡にて観察した。この手法を確立することはRNAウイルスであり、かつ感染量が少ないHCVの組織上での検出方法の開発のうえで重要と考えられる。
