本年度研究は計画通り順調に遂行され、以下の成果を得た。1.ヒト臍静脈内皮細胞、ウシ網膜周皮細胞を用いた低/無酸素(10%、5%、2.5%、0%O_2)培養系を確立した。2.内皮細胞、周皮細胞ともに低酸素下で生細胞数が有意に増加することが認められた。増殖促進における酸素の最大有効濃度は内皮細胞で10%、周皮細胞で2.5%であった。3.内皮細胞、周皮細胞の両者で酸素濃度の低下に伴いvascular endothelial growth factor(VEGF)遺伝子の発現が誘導された。4.内皮細胞はflt1,flt4、KDRの3種のVEGFレセプターをコードする遺伝子を酸素濃度の高低にかかわらず恒常的に発現していた。一方、周皮細胞ではflt1mRNAのみが低/無酸素状態で検出された。5.ヒトVEGF mRNAに相補的なホスホロチオエ-ト型アンチセンスDNA計11種を独自に設計し化学合成した。6.アンチセンスDNAでVEGF遺伝子の発現をブロックすると、10%酸素濃下での内皮細胞のDNA合成が抑制された。11種のアンチセンスVEGFDNA全てが有効に働き、対照のセンスDNAは影響をおよぼさなかった。このことは、VEGF遺伝子産物が内皮細胞の増殖に機能的にも関わっていることを示す。したがって、本研究により、低酸素による内皮細胞増殖誘導の少なくとも一部は「血管VEGF」のオートクリンおよびパラクリン作用を介するすることが明らかにされた。
