本研究の目的は生物学的薬物体内伝達媒体治療(biological drug delivery system:BDD)の動脈硬化治療応用への可能性を検討することにある。研究は(1)培養したウサギ大動脈血管内皮細胞を標識して動脈内注入にて末梢血管床への生着実験、(2)β-galactosidase分泌能を遺伝子工学的に組み込んだウサギ血管内皮細胞が末梢血管床生着後に局所組織でのβ-galactosidase活性を組織化学学染色した。ウサギ5頭から採取した培養血管内皮細胞を^<75>Seで標識し腹部大動脈より開腹下に注入したところ、最大8時間まで両側下肢より放射活性が検出されたことから末梢血管床に短時間は留まることが判明した。しかし、標識細胞が生着し増殖するのか、退縮するのかはこの実験系からは分からず今後の課題となった。また、両上肢からも弱い放射活性を検出したことから、培養細胞が末梢血管床を通過して全身循環へ再分布した可能性がありBDDの安全性も検討する必要があった。別のウサギ5頭には同様に腹部大動脈よりβ-galactosidase活性を有する培養内皮細胞を注入した。しかし4頭は実験経過中に細菌混入した可能性があり4頭とも敗血症で失い結果は得られなかった。1頭は3カ月後に犠牲死させて両側下肢の動脈と筋組織を採取して組織化学染色したがβ-galactosidase活性は得られなかった。培養細胞が退縮または生着するが分泌活性能を失うのか検討が必要であった。
