光合成は、ピコ秒レベルの早い光化学反応から、還元力とATP生成のための電子伝達、またそれらを利用したCO_2,N,S等の無機物の還元同化まで一貫した反応と捉らえる必要がある。さらにこれらの反応は環境から幾多のストレスを受け、適応しながら植物は生育して行く。このように植物の光合成には4つの調節機構がある。1.2つの光化学系レベルでの光エネルギー補集の調節2.循環的電子伝達系の制御3.ATP生成とNADPH生成のバランス4.還元型フェレドキシンの還元力が、NADP還元、無機物の還元同化、および酵活性の調節を行うフェレドキシン-チオレドキシン系にどのように配分されるか本年は国内のエキスパートの外、アリゾナ州立大学(テンピ)のブランケンシップ教授、ネブラスカ大学(リンカーン)のゴルベック教授およびカリフォルニア大学(バークレー)のブキャナン教授を加え、共同研究を組織する。星名は従前からポリエチレン・グルコールと熱処理を利用し、ホウレンソウ葉緑体から調製した光化学系I複合体を更に単純化し、反応中心の2つの大サブユニットの外、センターABを保持するタンパク質および1〜2個のタンパク質から成り、わずかなアンテナ・クロロフィルを有する複合体を用いてP_<700>からセンターXへの、またセンターXからセンターABへの電子伝達を測定し、速度論的解析を行った。大岡は、緑色イオウ細菌の光化学系を調製することに成功し、その構成成分を解析すると共にそれらの電子伝達反応をアリゾナ州立大学(テンピ)のブランケンシップ教授との共同で研究した。電子伝達反応の解析からも構成成分の存在を明らかにすることができ、緑色イオウ細菌の光化学系は典型的なFe-Sタイプの光化学系であることを確認した。この系は、大量に調整することができれば、X線による結晶構造解析の可能性があり、非常に興味深い。佐伯は、細菌におけるフェレドキシンを中心とした電子伝達の競争に遺伝子の改変によりタンパク質に変化を導入したものがどのようなeffectsを及ぼすのかをフィラデルフィアのペンシルベニア大学大西智子氏およびダルダル氏の協力で検討した。さらに光合成細菌のフェレドキシンを大腸菌中で発現させ、N_2固定との間の電子伝達がどのように調節されるのかを検討するものである。同様にゴルベックは光化学系I反応中心複合体を構成する種々のサブユニット・タンパク質遺伝子を改変し、複合体自体のアッセンブリーと遺伝子改変の影響を調べた。多くのものは複合体のアッセンブリー自体を防げるようであるが、数種のものは非常に興味深い結果となっている。非光合成組織のplastidsを利用した電子伝達機構の調節の解析については和田、松原、ブキャナンが協力して研究し、ホウレンソウの根およびモヤシマメの幼植物を用いて、フェレドキシンとフェレドキシン-NADP還元酵素(FNR)を精製し、詳しい性質を調べた。モヤシマメの幼植物の葉緑体には通常非光合成組織でのみ発現しているroot型ののFNRが発現していることが明らかになり、その生理的性質から非光合成的に電子伝達を行い、Nの代謝に関係していることが示唆され、Plant Phyhsiologyに投稿することができた。すなわち、幼植物では暗所でも貯蔵されている糖を利用して、酸化的pentose phosphate pathwayから生成されたNADP-FNRを介してフェレドキシンを還元し、窒素の代謝を行っているようである。これはやがて光合成能力の増大と共に失われ、通常の葉緑体の機能となるらしい。また、トマト果実の熟成に伴う葉緑体からchromoplastへのplastid変換についても、順調に進行している。ブキャナンはフェレドキシン-チオレドキシン系を非光合成組織に拡大し、いろいろの実用的な利用法を検討した。ブランケンシップ氏およびゴルベック氏が同時に日本へ招聘できたことによって、Fe-Sタイプ光化学系反応中心に関する小集会を開催し、全国から約10名の関連研究者が集まり、討論した。
Regulation mechanism of the electron transport in chloroplast and the related plastids was investigated by the group including three American scientists who was experts in each field. Experiments were planed by focussing on 1) regulation in cyclic electron transport system, 2) regulation of photon cupturation by two photosystems, 3) balance between NADP photoreduction and photophosphorylation, 4) partitioning of electrons from reduced ferredoxin to electron requiring system, such as NADP reduction, reductive assimilation of nitrogen, sulfur and carbon, ferredoxin-dependent thioredoxin system.Electron transport in photosystem I reaction center complex and Fe-S type reaction center was intensively investigated by spectrophotometrical and biochemical procedures and site-directed mutagenesis of genes encoding subunit proteins of the complex. It was investigated if the c-type cytochrome deficient mutant of photosynthetic bacterium (Rhodobacter capsulata) was compensated by the introduction of gene encoding human cytochrome c. At the moment no positive result was obtained.Some proteins components (ferredoxin and ferredoxin-NADP^+ oxido-reductase) of electron transport system in non-photosynthetic plastids were biochemically compared with those in chloroplast. There were isoenzymes or isoproteins differently expressed in chloroplast and non-photosynthetic plastids, which can differently function in the various electron transport systems in plastids.
