本研究では,覚醒剤であるメタンフェタミンやフェンテルミン,エフェドリン及び2-フェネチルアミンについて,迅速・正確な分析方法の開発を目指し,血清,尿や水に標準品を添加した試料から分析を試みた。方法はこれまで揮発性物質の分析に用いてきた,キーリーポイント・パイロライザー(熱分解器,Py)を使用し分析を試みた。まず,パイロライザーの至適条件を決めるために,n-プロピルホルミル及びイソブチルホルミル誘導体化した試料を用いてヘキサンにて抽出後,分離カラムとしてDB-17ワイドボア-カラム(0.53mmX15mX1.0μm)を使用したガスクロマトグラフィー・質量分析(GC-MS)を行ったところ,パルス温度は220-315℃で,また,パルス時間は4-6秒で良好なピークが検出された。以後,パルス温度は255℃,パルス時間は4秒で分析を行った。上記誘導体化薬物はカラム温度(100℃→240℃;16℃/分)の条件で良好に分離され,それらの検出限界は約0.3-0.6ng/インジェクションであった。更に,1.5-30ngの範囲で薬物濃度とマスクロマトグラムのピーク面積の間に直線関係が認められた。血漿や尿に上記薬物を各1μg/ml添加し、回収率を測定したところ71-78%の範囲であった。上記Py-GC-MS法による全分析所要時間は30分であった。以上の結果から,本法は覚醒剤類の迅速分析に有用であり,法医中毒学的実務に十分応用できることが明らかとなった。
