脳特異的な細胞外マトリックスとmatrix metalloproteinasc=MMPとの相互作用調べる中で,脳内に存在するプロテオグリカン(PG)であるtestican familyが細胞膜貫通型MMP(MT1-MMP,MT3-MMP)を抑制することにより神経膠芽腫細胞の浸潤を抑制することが明らかとなった。次いで正常脳及び浸潤能の著しい神経膠芽腫組織よりRNAを抽出し,testican familyの発現量を検討したところ正常脳では高発現を認め,腫瘍組織で発現の低下が認められた。更にtestican familyの発現ベクターを作成し,様々なinvasion assayでtestican familyの浸潤抑制効果を検討している。一方で近年単離された細胞外マトリックス分解酵素であるアグリカネース-1,2を含むADAMTS (A Disintegrin and Metalloproteinase with Thrombospondin motifs) familyの神経膠腫における発現についての検討を行い,アグリカネース-2が腫瘍の組織学的悪性度を反映することが示された。アグリカネースは既に正常脳で発現することが知られており,脳固有のPG分解に関係することが示唆されている。現在アグリカネース-2の発現ベクターを作成し神経膠腫細胞株への遺伝子導入を行って浸潤能に与える影響を検討している。今後はin situ hybridization法にて組織内での局在を,また遺伝子導入した神経膠腫細胞株の実験動物への移植を計画している。PGに富んだ脳組織の中で,ある種のPGは浸潤を抑える作用があることが明らかになり,また脳内PGを特異的に分解する酵素については現在まで未知である。我々の研究は脳腫瘍の浸潤機構を解明する上で細胞外マトリックスとその分解酵素の観点から新たな知見を得ようとするものである。
