組織・細胞に特異的な遺伝子の発現を組織標本上で可視化して検出する場合、通常行われる遺伝子組織化学(in situハイブリダイゼーション)法では、特定のmRNAの配列に相補的なDNAまたはRNAを放射性アイソトープ(RI)や非RI性マーカーで標識してプローブとし、組織内のmRNAと交雑させる。この場合、検出されるシグナルはその時点で細胞質内にすでに存在するmRNAの量を反映し、必ずしもその時点でDNAから転写されるmRNA量を反映するものではない。本研究ではmRNAの側を標識した新しい遺伝子組織化学法の開発を試みた。培養3T3-L1細胞において、短時間に転写される全RNAを^3H-ウリジンでRI標識した後に固定し,28SrRNAに対する非RI標識リボプローブを用いたin situハイブリダイゼーションを行い、交雑しなかったRNAをRNaseにより分解してからオートラジオグラフィーで解析したところ,アンチセンスプローブによる反応でセンスプローブによる反応より有意に多くの標識が核に残存した。同様な解析を,脂肪細胞に転化した3T3-L1細胞においてグリセロール-3-リン酸脱水素酵素のmRNAに対するリボプローブを用いて行ったところ,アンチセンスプローブによる反応でセンスプローブによる反応より有意に多くの核標識が得られた。脂肪転化しない細胞では両プローブによる反応の差は見られなかった。この結果は、ある時点における特定の細胞による特定の遺伝子の転写を組織標本上で検出できる可能性を示した。本法はRNAseプロテクション法の組織標本への応用といえる新しい組織化学技術であるので、「in situ RNAseプロテクション法」と名付けた。
