RNAポリメラーゼII最大サブユニットC-末端領域(CTD)が、リン酸化されることによって様々なmRNAプロセシング因子と特異的に相互作用出来ることが近年明らかになり、転写とmRNAプロセシングは、有機的に連携しながら進行している過程であると考えられるようになって来ている。本研究は、転写装置とmRNAプロセシング装置との間にどのような分子間相互作用のネットワークが存在し、転写反応及びmRNAプロセシング反応がどのように相互に関連し制御されているのかを明らかにする手がかりを得るために、リン酸化CTDに結合する新規蛋白質の同定とその機能解析を行うことを目的としている。これまでに、Far-western法及びGST-pull down法等によって、リン酸化CTDに結合する蛋白質として、ヒトの新規蛋白質PCIF1、既知のヒト蛋白質でペプチジルイソメラーゼ活性を有するPin1、mRNAスプライシング因子と考えられるFBP11及びHECTドメインを持ちユビキチンリガーゼ活性を有すると予想されるWWP1を同定した。これらの蛋白質が共通して持つWWドメインが、リン酸化CTDとの結合に必須であること、更にPCIF1については、リン酸化RNAポリメラーゼIIとの細胞内会合を示唆する結果を得た。現在これらリン酸化CTD結合蛋白質の細胞内機能の検索を行っている。PCIF1及びPin1を各々数種のヒト培養細胞に於いて一過性に過剰発現させたところ、いくつかの転写因子の転写活性化ドメインに依存したレポーター遺伝子のトランス活性化が、PCIF1及びPin1の発現量に相関して強く抑制されることが観察された。PCIF1及びPin1が、リン酸化RNAポリメラーゼIIを標的とする新たな転写抑制因子として機能している可能性が考えられる。
