ラットC6 glioma cell lineに対して、Fas抗体(IgM)を加え、Cell deathの形態を観察した。Fas抗体は30ng/ml,50ng/mlの濃度で培養液に添加した。コントロールと添加直後、24時間後、48時間後、72時間後、96時間後に光顕的、電顕的、ならびにin situ nick-endolabelling法で観察した。培養細胞は浮遊したものも採取し、遠沈後、光顕的には4%PFAで固定し凍結包埋した。電顕的には2%GAで前固定し、Osmiumで後固定しepon包埋した。また、ラットの脳内にmicroinjectorでFasを注入しラット脳の変化を観察した。C6培養細胞では、Fas添加24時間後から細胞縮小が始まり、72時間後には多数のearly apoptosisの所見を得た。96時間後には、apoptotic bodyが見られた。in situ nick end-labelling法ではapoptosisに陥った核に陽性反応がみられた。免疫組織化学的にC6にFasは発現していたが、Bcl-2蛋白は発現していなかった。ラット脳において、Bcl-2は神経細胞、microglia、perivascular cellに発現がみられた。Fasは神経細胞と一部のmicrogliaに見られた。Fas注入ラットは、1ヵ月生存しており、注入部分の神経細胞死はみられなっかた。以上により、Fasはラット悪性神経膠腫の培養細胞に有効であり、ラットの正常脳にはほとんど影響がなかった。また、神経細胞もFasを有しているがBcl-2を同時に有することにより、Fasによる細胞死(apoptosis)をまぬがれると考えられた。
