Fdを中心とする電子伝達系は葉緑体をはじめ、種々のプラスチド中でも重要な役割をもっていることは、多くの知るところである。しかしながら、FdやFNRのイソ酵素(タンパク質)の存在が明らかにされてから、このようなイソ酵素(タンパク質)の発現様式や性質さらにはそれらの機能は何かという新たな問題が生じて来た。このような背景の下に計画された本研究は、根、葉、果実などの器官のプラスチドに特異的に発現するFdとFNRを分離・精製し、それらの性質・分布・発現を明らかにした。Fdに関してはトマトの果実の成熟過程における葉緑体からクロモプラストへの変換に伴う時期的・組織的な特異的発現を明らかにすることができた。新たに果実でのみ発現するイソタンパク質の存在も明らかにした。またFNRについては葉および根から調製された標品で互いにはっきりと区別可能な特異的抗体を作成し、光合成タイプと非光合成タイプの組織でそれぞれのイソ酵素がどのように分布するかを明らかにした。特異的抗体を利用して進化的にいつごろ根のFNRと葉のFNRとが分岐したかを推定することができた。すなわち真の根器官が現れるコケ植物またはシダ植物の出現のころに遺伝子が分化したと考えるとうまく説明ができる。機能については、光合成能のない器官では酸化的ペントースリン酸経路で作られるNADPHからFNRによってFdを還元し、Fd依存性の種々の還元酵素への電子の供給を行っていることが明らかとなった。さらに、ホウレンソウ根から非光合成器官で特異的に発現するFNR遺伝子のクローニングを試み、ネステッドPCR法によって成功した。これによって根と葉のFNRの一次構造上での比較によって、どの部分が逆方向の電子伝達に重要なのか、今後、各器官での特異的発現を明らかにする準備が整ったと言える。
We have investigated characterization, function and expression of ferredoxin isoforms and ferredoxin-NADP^+ oxidoreductase isoenzymes specifically expressed in the various plastids in higher plants. We obtained the following results for three years.1) Purification, sequence determination and comparison of ferredoxin isoforms2) Characterization of leaf-type and root-type of FNRs3) Distribution and differential expression of FNR isoenzymes4) cDNA cloning of root-type FNR of spinach5) Estimation of divergence time of leaf-and root-type FNRs6) Function of these isoforms in nonphotosynthetic tissues
