• Note (General): 金沢大学がん研究所; C4低産生系マウス(H-2^k由来)では、C4遺伝子の第13イントロンに、レトロポゾンの一種、B2反復配列が挿入されていることを報告したが、本研究で、実際にこの異常挿入配列がRNAスプライシングの異常をもたらして正常C4mRNAの肝臓での恒常レベルを低下させる唯一の原因であることを、リコンビナントC4遺伝子の培養細胞へのトランスフェクションの実験によって証明した。また、B2反復配列挿入遺伝子の発現が組織特異的にも調節されていることも明らかにした。リコンビナントC4遺伝子は、C4低産生系と高産生系のマウスのC4遺伝子の第13イントロンをお互いに入れ替えて作成しそれらをプラスミドPUC19にサブクローニングしてHepG2細胞に導入した。HepG2細胞中のマウスC4mRNAを逆転写-PCR法とRNase Protection法で解析すると、B2反復配列が挿入されたリコンビナントC4遺伝子が導入された場合のみ、異常C4mRNAの出現と正常C4mRNAの低下が認められた。C4低産生系マウスでも、腹腔マクロファージでは、C4産生は低下せず、異常C4mRNAは検出されなかった。しかしながら、リコンビナントC4遺伝子を種々のマウスマクロファージ由来培養細胞に導入する実験を行ったところ、得られるC4mRNAは異常なスプライシングを受けたものが多く、正常C4mRNAは低下していた。; A retroposon B2 sequence is inserted into the 13 intron of the complement C4 gene of H-2^k derivred C4 strains that are characterized by the low C4 production. Aberrantly processed C4 mRNA and decreased level of C4 mRNA are detected in these mouse strains. To test the possible causal relationship between the B2 insertion and low C4 production in H-2^k derived strains, we constructed recombinant C4 gene with or without the B2 sequence and transfected them into cultured hepatoma cells. The transfection of recombinant C4 gene without the B2 sequence always resulted in the production of only normal C4 mRNA at the normal level, while transfection of recombinant C4 gene with the B2 sequence resulted in the abnormal splicing of C4 gene transcript and devreased level of normal C4 mRNA.Tissue-specific regulation of RNA processing for C4 gene transcript in H-2^k mouse strains are identified by analyzing the C4 mRNA from various tissues.
