1 マウス補体第4成分(C4)遺伝子群の転写調節:マウスの重複C4遺伝子、C4遺伝子とSex-limited protein(Slp)遺伝子は、95%以上の高い相同性を示すのに、その発現には著しい違いがある。C4遺伝子が構成的に発現するのにSlp遺伝子はアンドロゲンに誘導されてはじめて発現するのである。われわれは両遺伝子の転写活性に大きな違いがあることを見いだした。両遺伝子の5'上流領域から種々の長さのフラグメントを切り出してきてCATアッセイを行ってC4遺伝子では転写開始点と-380の間に強いプロモーター活性があるのにSlp遺伝子ではほとんど認められなかったのである。C4遺伝子ではこのプロモーター領域にNF・kB部位、NF1部位が存在するがSlp遺伝子では対応する部位には複数の変異が見られ、HepG2核抽出物とは結合しなかった。in Vitro mutagenesisによってそれぞれの部位に変異を与えたプロモーター領域を用いて、CATアッセイを行った結果、C4とSlpの転写活性の違いはNF・kBではなくNF1で大部分説明されることが明らかとなった。2 ヤツメウナギからの補体B因子と補体第2成分の祖系遺伝子の分離:哺乳動物のB因子とC2に共通に保存されている配列をプローブとして用い、現存する最も原始的な脊椎動物種である円口類ヤツメウナギの肝臓から、B因子とC2の祖系遺伝子を分離した。cDNAクローンは2。6kb長で、哺乳動物のB因子、C2と同様に3種のドメイン(SCR反復配列、von Willebrand,セリンプロテアーゼ)からなるモザイク蛋白をコードしていた。蛋白全体としてのアミノ酸配列上の相同性は、マウスB因子に対しては33%、マウスC2に対しては29%であった。
