1)抗タグ抗体を用いた複合体精製による相互作用の確認、プロファイリングおよび複合体全体の構成解明を効率的に推進する為に、新規タンデムアフィニティ精製(TAP)タグの開発を行った。変性剤存在下での精製に適用可能なHis6に加えてStrept-tagとFlag-tagを様々な順列で配置したタグを作製し、テスト蛋白質の精製効率から性質のよいものを選択した。これを用いて酵母の全SH3蛋白質遺伝子にタグコード配列をノックインした株を作製、架橋剤存在下での複合体精製とLC/MS/MSによる解析を進めている。2)異なるタグで標識されたユビキチンを同時に発現し、双方のタグに対するアフィニティ精製を連続して行うことで、ポリユビキチン化蛋白質を高純度に精製するパラレルアフィニティタグ精製(PAP)法を開発し、得られた蛋白質のLC/MS/MSによる網羅的同定を行った。同定された蛋白質に関しては個別にユビキチン化を検討して、この方法による検出の正当性を実証した。3)保証付き逆2ハイブリッド法による相互作用変異体の単離法の開発と応用を進めた。その結果、これまで解析が難航していたほ乳類細胞極性蛋白質Par6とaPKCの相互作用や、出芽酵母の細胞極性蛋白質BEM1とCDC42の相互作用の解析に成功し、本法の有効性が実証された。前者は更に我々が見い出したPB1ドメインの認識機構の研究に、後者は新規CDC42結合ドメインの発見へと研究が展開している。これらの経験も踏まえて、保証付き逆2ハイブリッド法を用いて得られた相互作用変異体と同時に相互作用への中立変異体とを配列決定することで相互作用モチーフの同定が効率的に進展できる可能性が示された。
