Matrix metalloproteinase(MMP)遺伝子ファミリーは、一次構造と酵素活性が明らかにされた9種類の分子種から構成されている。本研究では、潜在型MMP-7(proMMP-7)を精製しその生化学的性質を検討して、以下の新しい知見を得た。1)ProMMP-7(分子量28,000)は、p-aminophenylmercuric acetate(APMA)で活性化され、分子量19,000の活性型に変換した。また、トリプシンでも活性化される他、プラスミンと好中球エラスターゼによっても全活性の50%まで活性化された。しかし、HOClやH_2O_2では全く活性化されなかった。2)ProMMP-7は、活性型MMP-1,2,3,9のうちMMP-3(モル比1:5)によってのみ特異的に活性化された。一方、MMP-7の潜在型MMP-1,2,3,9に対する作用を検討したところ、proMMP-1をAPMAによる活性化と同レベルにまで活性化し、APMA共存下ではその活性を約7倍“superactivate"した。また、proMMP-9もMMP-7(モル比1:1)で50%まで活性化された。しかし、潜在型MMP-2とMMP-3はMMP-7で全く活性化されなかった。3)活性型MMP-7はプロテオグリカン、フィブロネクチン、ラミニン、ビトロネクチン、ゼラチン、IV型コラーゲンおよび不溶性エラスチンを分解した。ことにエラスチンに対してはMMP-1,2,3,7,9のうち最も強い活性を示した。以上のデータは、MMP-7が種々のセリンプロテアーゼや他のMMPとの相互作用の結果、炎症や癌の浸潤・転移の際の細胞外マトリックス分解に作用しうることを示唆している。これらの成果に対して現在論文執筆中である。慢性関節リウマチ関節組織内でのMMP-7の局在に関しては、合成ペプチドに対するモノクローナル抗体がMMP-1と交差反応したため十分なデータは得られなかった。現在、精製したproMMP-7を抗原として特異的モノクローナル抗体の作製を行っており、今後検討する予定である。
