本研究は、腫瘍発生、悪性化メカニズムを幹細胞の分化や未分化性維持機構との関連を中心に解き明かそうというコンセプトに基づいた萌芽的研究である。具体的には、悪性脳腫瘍であるグリオーマをモデルとして、悪性度の進展における未分化性獲得メカニズムの解明に取り組んだ。グリオーマgrade IIIとgrade IVの違いを詳細に比較検討することによって、腫瘍発生・悪性度進展メカニズムの解明を目的に、神経幹細胞におけるEGF-RAS経路活性化状態における細胞分化運命決定に対する影響を解析した。その結果、RAS活性化が神経幹細胞のグリア系への分化を誘導すること、この分化誘導が阻害されると腫瘍形成が促進されることを見い出した。さらに、本モデルを遺伝子改変マウスと組み合わせることによって、ヒトのグリーマgrade IIIあるいはgrade IVと同じ病理的特徴を持つ腫瘍を再現させることに成功した。悪性度の高い腫瘍(GBM,grade IV)ほど、未分化抗原の発現が見られ、一方悪性度の低い腫瘍(Anaplastic astrocytoma,grade III)では、分化抗原が高い発現を示していた。grade IVはsphere形成能を示すが、grade IIIではその活性が見られないなど、悪性度と未分化性の相関を確認した。この研究成果は、脳腫瘍悪性進展に関わる因子の同定に寄与すると考えられ、予後因子の特定を含む診断技術の向上、新規治療法の開発を目指す礎となる。
