Note (General): 金沢大学薬学部
分裂酵母のweel変異を相補できるヒトweel遺伝子産物を大腸菌内で発現精製し、その酵素活性を検討した。その結果ヒトweelキナーゼはサイクリンB/cdc2キナーゼのTyr-15を燐酸化しその活性を阻害することを明らかにした。しかしこの酵素はThr-14を燐酸化できなかった。このことはThr-14を燐酸化しp34^<cdc2>キナーゼ活性を阻害する新たなキナーゼが存在することを示唆している。一方、3種のヒトcdc25フォスファターゼを大腸菌内で発現させ、これら酵素の活性も検討した。前記したp34^<cdc2>キナーゼの燐酸化による活性阻害はこのヒトcdc25フォスファーターゼにより回復する。3種存在するヒトcdc25フォスファターゼのうち少なくともcdc25BはこのTyr-15のみならずThr-14の燐酸基も脱燐酸化し、その活性を上昇させる。さらにcdk2の活性制御に関連してこの酸素の燐酸化部位を検討したところTyr-15とThr-160が確認された。このThr-160が燐酸化されるとSDS電気泳動で移動度が速くなり、これは蛋白質、p34^<cdc2>キナーゼを含め、一般に、燐酸化されると移動度が遅くなる現象とは異なる。サイクリンA結合cdk2はこのThr-160の燐酸化体であったことから、サイクリン結合にはこのThr-160の燐酸化が必要であると考えられた。サイクリンA/cdk2のTyr-15の燐酸はヒトCDC25フォスファターゼではサイクリンB/cdc2キナーゼのようには効率よく脱燐酸化されず、この原因がサイクリン蛋白質の違いによるのかどうか、Tyr-15の生理的意味を含めて興味深い。
