<神経発生学的研究>BrdUの免疫組織化学で、特定の胎生期に分裂発生したニューロンを成体で同定することに成功した。神経トレーサーである蛍光色素で標識された神経細胞と二重標識して観察する手法も我々が独自に開発して、国際学術誌に公表した(Kitao et al.,Brain Research、1993年)。上オリーブ核-下丘投射ニューロンのうち、交叉性投射ニューロンは発生時期が早く(E12-E13)、非交叉性投射ニューロンは発生時期が遅い(E14-E16)ことを世界ではじめて発見した。投射の側性(laterality)によってそのニューロン群の発生時期が異なるという仮説が本研究ではじめて証明された。国際学会で発表(International Workshop on Binaural Processing in the Auditory Brainstem.Ottawa,Canada,1993年6月;第16回日本神経科学学会.大阪、1992年12月;Society for Neuroscience.Anaheim,California U.S.A.,1992年10月)。<脳幹神経回路網の生後発達;脳損傷の影響とその修復および代償過程>内側上オリーブ核-下丘投射の発達は短い期間に成立するものではなく、それらのニューロンのなかには、生後発育中の様々な時期に軸索を下丘まで伸展していくものが混在していることが判明した。新生児期に一側下丘を実験的に剥離した場合の成長後の変化を調べた結果、逆行性の細胞死、異常な線維連絡の形成、投射標的の剥離に対する抵抗性の出現等の興味ある現象がおこることが証明された。第98回日本解剖学会総会(札幌、1993年7月)で口頭発表され、原著論文に公表した(尾小山1994年)。
<Neurogenetical study>By a combination of fluorescent retrograde axonal tracings and bromodeoxyuridine (BrdU) immunohistochemical techniques, time origins of projection neurons in the superior olive have been examined. In the lateral superior olive, neurons projecting to the contralateral inferior colliculus (IC) were generated on days E12 and E13, whereas neurons projcting to the ipsilateral IC were proliferated between days E14 and E16. In the medial superior olive (MSO), the IC projection neurons arose on E12, while, in the superior paraolivary nucleus, their peak production occurred on day E13. Also refer some publication (Kitao et al., brain Research, 1993) and oral presentations (International Workshopon Binaural Procssing in the Auditory Brainstem. Ottawa, Canada, 1993 ; Society for Neuroscience.Anaheim, California U.S.A., 1992).<Normal development of auditory brainstem pathways and plasticity after unilateral ablation of the IC>The results indicated that : 1) The projection froj the MSO to the IC was developed gradually in postnatal periodsas an exclusively uncrossed one without making axon-collaterals to the contralateral IC.2) When the IC was ablated unilaterally in certain postnatal period, many neurons died in the ipsilateral MSO as a result of axotomy. 3) An aberrant crossed projection to the contralateral IC occurs in some of the survived neurons in the MSO, although such aberrant fibers still maintained in adult were very few in number. 4) Growth of late-developing axons was a major factor in the plasticity of this system of projection.
