- 2009-2011年度科学研究費助成事業(科学研究費補助金(基盤研究(C)))研究成果報告書 課題番号：21580350 研究代表者：佐藤正宏 (鹿児島大学フロンティアサイエンス研究推進センター教授)本研究ではどんなタイプの遺伝子でもその遺伝子発現をノックアウトできるブタ細胞における汎用性の高いdouble KO細胞を作成する方法を開発することを目指した。具体的には、ヘテロKO細胞targeting vector （TV）を再度導入し、薬剤濃度を数倍高めた条件で選別する方法（repeated delivery of TV, RDTV）を用い、特定の遺伝子の発現を抑えることに成功した。This study aims to develop a general method for obtaining double KO swine cels in which the target gene expression is completely blocked. This was successfully performed by selection of heterogenous KO cels under higher concentrations of selective drugs after repeated delivery of a targeting vector (RDTV). These resulting double KO cells will be used for somatic cell nuclear transfer-mediated production of cloned pigs.
