本研究では、口腔癌におけるMUC1遺伝子のスプライシング異常の検出を行い、臨床病理学的事項との関連について明らかにすることを目的とした。まず、口腔癌症例の組織を用いて膜型ムチンの発現を検索し、MUC1およびMUC4の過剰発現は、口腔癌の有意な予後予測因子であることを明らかにした。その後細胞株や切除組織を用いて、MUC1のスプライシングバリアントに対し個別の検討を開始した。その結果、それぞれの細胞株や症例により、発現バリアントの種類およびその比率は大きな相違があることが明らかになった。
The goal of this study was to detect alternative splicing of MUC1 mucin in oral squamous cell carcinoma and to evaluate its relationship with clinicopathological factors. First, we investigate the expression level of membranous mucin in the resected tumor samples by immunohistochemistry and found that aberrant expression MUC1 and MUC4 was an independent prognostic factor indicating poor prognosis in patients with OSCC. Next, we have started to evaluate the expression level of each splicing variant of MUC1, using the specimen from surgically resected oral squamous cell carcinoma. The expression level and proportion of MUC1 splicing variants showed a huge variety in each specimen.
