- 2009-2011年度科学研究費助成事業(科学研究費補助金(基盤研究(C)))研究成果報告書 課題番号:21590341 研究代表者:岸田昭世 (鹿児島大学大学院医歯学総合研究科教授)ヒトグリオーマ(神経膠腫)由来の細胞株におけるすべてのWntファミリーの発現を解析し、Wnt-5Aの有意な発現の亢進を認め、Wnt-5A発現亢進がグリオーマの細胞運動、MMP-2発現調節を解して浸潤能を調節していることを見出した。また、がん化に関わる可能性があるHPVやSV40ウイルスを使わずにヒト正常口腔上皮由来細胞MOE1の樹立に成功し、これらの細胞株でWnt-5Aが発現していることを見出した。以上のことから鑑みて、当初の計画以上に進展していると判断している。We analyzed the expressions of all Wnts and Frizzleds in several human glioma cell lines and found that Wnt-5A, Wnt-7A, Frizzled-2, and Frizzled-6 were highly expressed in those cells. By Wnt-5A-knockdown experiment, we have shown that Wnt-5A stimulated induced cellular motility and invasion of human glioma cells through the induction of MMP-2 mRNA expression and MMP-2 enzymatic activity. We established normal human epithelial cells without using HPV and SV40 genes, which have the potential to induce carcinogenesis.
