多機能神経ペプチドPituitary adenylate cyclase-activating polypeptide (PACAP)は、精巣に高濃度に存在し、PACAPおよびそのレセプター発現が精子形成のステージにより周期的に変化する事から、生殖への関与が示唆されている。我々は、ヒトPACAP遺伝子の精巣特異的発現調節機構を目指し、精巣特異的エクソン上流に精巣特異的プロモーター領域を同定した。さらにこの領域に特異的に結合するタンパク質として、PARP-1とTIARを同定した。これらタンパクをsiRNAでノックダウンすると、80bpのプロモーター活性が変化しPACAPの精巣特異的機能への関与が示唆された。
Pleiotrophic neuropeptide, pituitary adenylate cyclase activating polypeptide (PACAP) is abundantly localized in testis. It was reported that gene expression of PACAP and its specific receptor PAC1 are periodically changed according to stage of spermatogenesis, suggesting its involvement in gonadal function. We attempted to clarify the testis specific regulatory mechanism of PACAP gene expression, and identified the promoter region responsible for testicular specific expression in 5’ upstream region of human testicular specific exon. In addition, we also identified PARP-1 and TIAR as a protein which specifically bind this region. Knock down of them with specific siRNA changed the testis specific promoter activity, suggesting that these proteins might be involved in testis specific function of PACAP.
