- 2011-2012年度科学研究費助成事業(科学研究費補助金(若手研究(B))研究成果報告書 課題番号:23760752 研究代表者:武井孝行(鹿児島大学・大学院理工学研究科・准教授)ポリメタクリル酸メチル(PMMA)からなる直径10μm程度のファイバーを調製した。そのファイバーの束をアガロースゲル内に包埋し、ファイバーのみを特異的に溶解することで、ゲル内に毛細血管網様流路ネットワークを作製した。続いて、多孔質化したそのゲルに肝癌細胞を播種後、その多孔質体をアガロースゲルで密閉した。多孔質体の流路内部に培地を流通させながら培養することで、培地を流通させなかった条件よりも、より多くの生細胞を多孔質体内部に保持できることが示唆された。Poly(methyl methacrylate)(PMMA)microfibers of approximately 10 μm in diameter were prepared using wet spinning technique. The microfibers were embedded in agarose gel and dissolved specifically, resulting in formation of capillary-like network in the gel. The gel was freeze-dried and hepatoblastoma was seeded into the sponge-like gel. Then, the sponge was coated with agarose gel. Culture medium was perfused into the capillary-like network in the sponge. Albumin productivity of the sponge with perfusion of medium was higher than that without medium perfusion.
