- 2009-2010年度科学研究費補助金(若手研究(B))研究成果報告書 課題番号:21790999 研究代表者:児玉祐一 (鹿児島大学医学部・歯学部附属病院医員)再生不良性貧血の進行に血小板内のVEGFが関与していることを以前証明したが、今回VEGFの転写因子であるHIF-1αが再生不良性貧血の進行に関与しているかを評価した。再生不良性貧血患者の骨髄の有核細胞を用いてHIF-1αのmRNAの発現量を評価したが、急性に進行する患者と慢性に進行する患者では有意差はなかった。VEGFには別の因子が働いている可能性があることを示唆した。今後巨核球を用いた実験系を確立する予定である。We assessed the level of hypoxia-inducible factor-1alpha (HIF-1α), which is known to be a transcription factor of VEGF. The mRNA levels of HIF-1α in bone marrow mononuclear cells (BMMC) were measured by real-time PCR in 13 aplastic anemia (AA) patients. HIF-1α in chronic AA patients was not significantly greater than that in acute AA patients. These data indicated that a high level of HIF-1α in chronic AA does not cause the high level of VEGF in a single platelet in chronic AA. The mRNA of HIF-1α in megakaryocytes should be analyzed to better understand the role of HIF-1α in AA.
