本研究は、ポリアミン蓄積による細胞死のメカニズム解明を目指して、spermidine/spemine-N1-acetyltransferase(SSAT)の強力な阻害剤を設計・開発することを目的とした。実験計画に従い、1)SSATの活性測定法の開発と阻害効果を持つ化合物のスクリーニング、2)ポリアミンアナログの合成と培養細胞への効果、3)ポリアミン過剰蓄積の際の細胞容積変化と細胞内分布に関する研究を行い、以下の成果を得た。N^1-Dansylnorspermineを蛍光基質とするHPLCによるSSAT活性測定法が、組み換え体ヒトSSATを用いるSSAT阻害剤のスクリーニング法として有用であることを明らかにした。また、組み換え体ヒトSSATおよびSSATを薬物処理により誘導したHTC細胞の抽出液について、詳細に酵素反応を速度論的に解析し、生体試料中のSSATの高感度な活性測定条件を確立し、細胞試料へ応用可能であることを明らかにした。確立した活性測定法を用い、SSATの阻害剤をスクリーニングしたところ、これまで報告のある阻害剤の中で最も強力とされるdi(ethyl)norspermineよりも強力であり、polyamine oxidase(PAO)に対しても阻害効果を持つ化合物を見いだした。 SSATスクリーニングに用いた数多くの阻害剤について、培養細胞に対する効果を検討し、それらが細胞増殖抑制、アポトーシスの誘導に有効であることを明らかにした。薬物処理してspermidineを過剰蓄積したラット肝がん由来HTC細胞の細胞容積の変化およびポリアミン細胞内分布を、それぞれ、コールターカウンターおよび限外ろ過法により調べ、ポリアミン蓄積が細胞容積を縮小させるばかりでなくポリアミン細胞内分布にも影響することを示唆する結果を得た。
