本研究では、高病原性鳥インフルエンザA/H5N1に特有な白血球減少症の病理を再現する数値的な体内モデルの開発および病理シミュレーションを行った。最初、遅延微分方程式による数理モデルの開発を行った。モデルは白血病減少症を再現したが、硬い方程式系となったため、計算時間の点で流行伝搬モデルとの連成は困難であった。このため、次に、常微分方程式モデルを開発し、実験データによってモデルのパラメータ同定を行った。さらに、モデルを用いて治療の効果の評価もおこなった。常微分方程式系のモデルでは、流行伝搬モデルと連成した高速なシミュレーションが可能であった。 In this study, we developed a numerical within-host model and performed simulations to reproduce the pathology of leukopenia peculiar to highly pathogenic avian influenza A/H5N1.As a mathematical model,we first developed a model by delay differential equation system. Although leukopenia could be reproduced,the system became stiff.It was difficult to combine the within host model with a transmission model in a local community from the view point of computation time.Then,we next developed an ordinaly differential equation model and identified parameters using experimental data.Furthermore,the effectiveness of neuraminidase inhibitor therapy and immunoglobulins therapy were evaluated using the ordinary differential equation model. High-speed simulation coupled with transmission model becomes possible.
