Note (General): 黒縞カイコのヘテロ接合体(P(sup s)/P)の休眠卵に重粒子線(Ne、C)を照射すると、艀化幼虫の5齢期に黒い皮膚をバックに白斑を持つ蚕が出現する。この頻度はNeの照射線量、エネルギー(LET)に依存した。しかし、休眠期間において重粒子線の感受性が異なり、休眠が覚醒して胚発育が再開した2日目の卵において放射線に対する感受性が最も高い。Cイオンビームを卵中央部(卵黄)と卵背面部(胚)に直径250μmの範囲で局部照射すると、後者で体細胞突然変異発生した。さらに、重粒子線照射によるカイコ遺伝子発現への影響について検討した。すなわち、休眠覚醒した黒縞蚕卵の胚発育2日目にNeイオンビーム5.0Gyを照射し、照射後0、2、4、6日目の時期に生じる遺伝子発現への影響を検討した。蚕卵からTotal RNAを抽出し、RT-PCR-Differential Display法を用いてDNA断片を増幅させ、無照射区の増幅DNA断片との違いを電気泳動のバンドの検出パターンを比較することにより解析した。その結果、照射区と無照射区において数種のバンドの検出量に顕著な違いがみられ、重粒子線照射より蚕卵内において特異的に発現する遺伝子が存在することを推察した。以上の結果は、カイコの卵への重粒子線照射の影響を個体レベルでの形質発現と分子レベルでの遺伝子発現において検出できることを示している。
Sensitivity of Bombyx eggs to a heavy ion beam was estimated from the frequency of somatic mutation which was shown by the white spot (s) on the black integument of the fifth instar larvae. The number of larvae with the white spot has been clarified as index of the frequency of somatic mutation incidence. The frequency was the highest on the day two eggs after the resumption of embryogenesis in the diapause-terminated eggs. The somatic mutation was induced by the irradiation of Fe ion directly to embryo. Differential-Display-Reverse-Transcriptase-Polymerase-Chain-Reaction showed that the irradiation of Fe ion on the day two eggs of the resumption of embryogenesis induced influences of gene expression, suggesting that the influence of cosmic rays on the gene (s) could be estimated using Bombyx eggs.
