Note (General): 重力場における植物細胞の応答を調べる目的で微小重力下における植物細胞の成長ならびに分化を調べた。今回特に飛行実験用に培養容器を準備したが問題なく使用できることが確認できた。一方、短期間の培養ではあるが、異なる細胞、培地、重力場(微小重力場、1G)いずれの組み合せにおいても植物細胞は生育、分化することが認められた。しかし微小重力場で培養した試料、特にカルス増殖培地に置床した茎切片において地上対照区より顕著な生育のバラツキが認められ、また新鮮物重の増加量も微小重力場で培養した試料の方が劣る傾向が認められた。さらに、重小重力場では地上対照区で観測される複数の不定芽の形成は認められなかった。これらの結果は予想されない結果であり、再実験により確認される必要がある。微小重力下ならびに1G下で培養した組織および細胞内の微細構造を観察した結果、分化培地で培養した場合、微小重力場で再分化した茎頂は1G下で分化した茎頂よりもその発達がわるいことを認めた。また、この時の色素体(葉緑体)の発達は1G下の方がよい傾向がみられた。また細胞骨格を形成する微小管の発達も1G下の方が優れている傾向が認められた。一方、脱分化培地においても茎の側芽において微小重力下にあるものの方がより色素体の脱分化が進んでる傾向が認められた。1次ならびに2次代謝に関係する酵素の活性を測定した結果、微小重力下、再分化培地にて培養したカルスにおいてリグニン生合成の律速酵素であるコーヒー酸o-メチル基転移酵素の活性が地上対象区より著しく低いことを認めた。なお、微小重力下と1G下におけるタンパク質合成活性を35S-メチオニンで標識したのち2次元電気泳動により分離し比較したが、顕著な違いを検出することはできなかった。以上の結果は微小重力場が植物細胞の生長、分化に細胞レベルで影響していることを示していたが、微小重力の影響を知るためにはさらなる実験が必要と考えられた。
Plant cells' growth and differentiation under microgravity were investigated for understanding their response in gravitational field. A specially-devised culture vessel for this space flight was confirmed to work satisfactory. Plant cells grew and differentiated with any combination of different cells, medium, and gravitational fields (micro gravitational field and 1 G), although the culturing period was short. However, samples cultured in micro gravitational field, especially stem sections in callus proliferation medium, showed wider growth variation than ground control samples. The increase in fresh weight also tended to be lower in samples cultured in micro gravitational field. Multiple adventitious bud formation in the ground control samples was not observed in the samples in micro gravitational field. Because these results are unexpected, further experiments are necessary to confirm them. Observing the cultured tissues and intracellular fine structure under microgravity and 1 G showed that the shoot apex redifferentiated in micro gravitational field, when cultured in differentiation medium, grew less than that differentiated under 1 G. The growth of plastid (chlorophyll) and microtubule forming cytoskeleton tended to be higher under 1 G. In dedifferentiated medium, the plastid dedifferentiation in stems' lateral buds also tended to be higher under microgravity. Measuring enzyme activities related to primary and secondary metabolism showed remarkably reduced activity of caffeic acid o-transmethylase, a rate-limiting enzyme of lignin biosynthesis, in a callus cultured in redifferentiated medium under microgravity, compared with that of the ground control samples. In addition, protein synthesis activities under microgravity and 1 G were labeled with S-35-methionine, separated by two-dimensional electrophoresis, and compared. However, it showed no remarkable difference. These results suggest that micro gravitational field has effects on plant cells' growth and differentiation at cellular level. Further experiments are necessary to understand the details of microgravity effects.
