Note (General): 本研究では、これまで開発してきた4 塩基コドンなどを用いたタンパク質の部位特異的二重蛍光標識法をさらに発展させることで、タンパク質の立体構造やその変化をFRET により解析する手法の確立を行なった。実際に、マルトース結合タンパク質などについて基質結合に伴う立体構造変化のFRET を用いた検出や、二重標識に有用なN 末端特異的な非天然アミノ酸誘導体の導入法、高効率アンバーサプレッサーtRNA の開発などを達成した。 : In this study, FRET analysis of protein structures and structural changes were investigated through site-specific incorporation of fluorescent nonnatural amino acids in response to expanded genetic codes. Detection of structural changes of maltose-binding protein upon ligand-binding by FRET, N-terminal specific incorporation of nonnatural amino acid derivatives, and screening of highly active amber suppressor tRNAs were achieved.
