研究成果の概要（和文）：本研究では、プロスタグランジン類の生合成を司る酵素の機能と反応について検討した。（１）プロスタグランジンH2合成酵素(シクロオキシゲナーゼ)：本研究では、シクロオキシゲナーゼ２型の酵素(COX-2)によるアラキドン酸の水素引き抜きの段階では、活性化エネルギーが、基質の立体配座に依存することをQM/MM法で明らかにした。（２）リポカリン型プロスタグランジンD合成酵素：プロスタグランジンD合成酵素の反応について検討し、システイン残基のチオラートイオンがエンドペルオキシド酸素に攻撃する経路を見いだした。比較としてビリン還元酵素と基質複合体の結合状態を明らかにした。研究成果の概要（英文）：The present studies are aimed for the functions and reaction mechanisms of　prostaglandin synthases.　(1) Prostaglandin H2 synthase (cyclooxygenase): We investigated COX-2 catalyzed reaction of　arachidonic acid, and we found that the activation energies for the proton abstraction step depend　on the conformations of the substrate.　(2) Lipocalin-type prostaglandin D synthase: This synthase catalyzed isomerization of prostaglandin　H2, and we found a pathway which involves nucleophilic attack of the thiolate ion of cysteine　residue into an oxygen of endoperoxide. We also examined electronic and bonding states of bilin　reductase-substrate complex.
