並列タイトル等ERK1/2 シグナル伝達経路を介したヒト歯肉線維芽細胞におけるShikonin の生物学的効果
一般注記Shikonin is an active ingredient in Lithospermum erythrorhizon that promotes wound healing. We examined wound healing by shikonin using human gingival fibroblasts (hGF) collected from patients. We determined the optimal concentration from the result of cell proliferation and migration. And then we investigated the expression of VEGF, FN and Type I collagen. Moreover, we examined the phosphorylation of ERK 1/2 involved in wound healing. Finally, we performed all experiments using ERK 1/2 specific inhibitor (PD98059).Shikonin 1μM significantly promote cell proliferation and migration, shikonin promoted the expression of VEGF, FN and Type I collagen. Also, shikonin enhanced the activation of ERK 1/2 phosphorylation PD98059 suppressed the these effects of shikonin. In conclusion, wefound that shikonin might be useful in gingival wound healing.
