アルツハイマー病 (ア病) は認知症の最大の原因である。我々は、ア病患者後頭葉よりア病関連神経細胞死を制御するヒューマニン(Humanin, HN)を同定し、HNおよびその誘導体であるコリベリン(Colivelin, CLN)の神経保護作用を検討したところ、JAK2/STAT3経路の活性化によってア病関連神経細胞死を抑制することが判明した。本研究では、STAT3の不活性化がア病発症に関与するかを検討した。この結果、年齢依存的に海馬神経においてSTAT3が不活性化され、ア病患者ではSTAT3不活性化が著明であることを見いだした。STAT3不活性化の主な要因は脳内βアミロイド（Aβ）の増加であり、STAT3不活性化によりアセチルコリン作動性神経の機能が障害されることが明らかとなった。STAT3を標的とする新しいア病診断・治療薬の開発が期待される。
