真性赤血球増加症の原因遺伝子であるチロシンキナーゼJAK2の点変異体(V617F)が誘導するがん化シグナルの分子機構を解析し、発症機構を分子レベルで理解することを目的とした。JAK2変異体は転写因子STAT5の恒常的な活性化を誘導し、顕著な細胞増殖や腫瘍形成を誘導することを見出した。また、JAK2変異体の下流において、分裂期キナーゼAurora kinase Aは抗がん剤シスプラチンよるDNA損傷に対する耐性に寄与し、セリン・スレオニンキナーゼAktは、抗アポトーシス作用に寄与することを明らかにした。
