105例の食道扁平上皮癌切除検体に対してケモカインレセプターCCR7の免疫組織染色を行ったところ、CCR7陽性例26例(27%)はリンパ節転移と有意に相関し、術後5年生存率は有意に不良であった。ヒト食道癌細胞株におけるケモカインレセプターCCR7の発現を確認した。さらに細胞遊走の機能的解析を行ったところCCR7を発現する細胞株は対応するケモカインCCL21の投与により濃度依存性に遊走活性を示した。一方食道扁平上皮癌細胞株、食道癌切除検体ともにケモカインCXCL12を発現しており、食道癌切除検体におけるCXCL12陽性例は有意に予後不良であった。食道扁平上皮癌細胞株においてCXCR4発現株、CXCL12発現株を作製し、増殖速度を検討したところ、CXCL12とCXCR4は食道癌において双方が発現してautocrineにより悪性度を獲得している可能性が示唆された。食道扁平上皮癌細胞株においてCXCR4発現株、CXCL12発現株で抗HIV治療薬であるAMD3100投与により細胞増殖が有意に抑制された。
