われわれはこれまでの研究により、漢方薬十全大補湯が1型インターフェロンであるインターフェロンαの遺伝子発現経路に作用してインターフェロン産生を制御していることを見出した。本研究ではさらなる機序解明のため、大腸の固有粘膜層に存在するインターフェロン (IFN)産生細胞について明らかにするとともに、Toll Like Receptor (TLR) 4およびMyD88ノックアウトマウスを使用してそのメカニズムを解明する事を試みた。その結果、十全大補湯はTLR4依存かつMyD88非依存のシグナル経路に働きかける事が明らかとなった。また、MyD88ノックアウトマウスではI型インターフェロン関連遺伝子の定常状態における発現レベルが高くなっており、十全大補湯により発現レベルが減少する事が認められ、同様の現象が無菌 (GF)マウスでも認められた。さらに、それらの作用細胞は全て同じ単球系細胞であったが、既報のインターフェロン産生細胞とは異なることが示された。次に十全大補湯の感染防御能を確かめる目的でインフルエンザ感染実験を行ったが、対照群の補中益気湯と比べ、十分な抗インフルエンザ効果が認められなかった。マウスの系統やウイルスの特性によって十全大補湯の実験系に影響を及ぼす事が考えられた。また補中益気湯について、その感染防御メカニズムの一端を明らかにした。
