本研究ではTfh細胞を中心としてエフェクターTh細胞の分化におけるSOCS1の作用の解析を行った。抑制性T細胞であるTregは、SOCS1の欠損によりFoxp3を失いIFN-γやIL-17を産生するエフェクター細胞(exFoxp3細胞)に転換し、自己免疫様疾患を誘発することをつきとめた。このexFoxp3細胞ではSTAT1やSTAT3が過剰に活性化しており、IFN-γやSTAT1の更なる欠損によってexFoxp3細胞の出現は抑制された。よってSOCS1はTregにおいてFoxp3の安定性及びIFN-γへの耐性を付与し、exFoxp3への転換を抑えていることが明らかとなった。
